제목 Development of a protease assay method via engineering of procaspase-3 enzymes
초록 Proteases are implicated in an essential role of the biological processes including diseases development. In this study, a novel activity assay method for matrix metalloproteinases is established by using an engineered procaspase-3 by replacement of cleavage site in the procaspase-3 to recognize by interest protease. As a result, the engineered pro-caspase-3 can be purely activated by MMP-2 and the enzyme activity was increased 225.9-fold after proteolysis. The assay could be distinguished pico-molar range of MMP-2. By development of the high sensitive and simple assay method for protease activity, the assay is expected to be applied diagnosis for MMP2-associated diseases and easily adapted for other proteases.
