Glucose-sensitive liposomes incorporating Poly(N-isopropylacrylamide-co-methacrylic acid-co-octadecylacrylate) and Glucose Oxidase |

We prepared the egg phosphatidylcholine and dioleoylphosphatidylethanolamine liposomes modified with various ratio of P(N-isopropylacrylamide-co-methacrylic acid-co-octadecylacrylate) (P(NIPAM-co-MAA-co-ODA)) and glucose oxidase (GOx). Copolymer incorporated liposomes exhibit significant release in the acidic conditions because of MAA, pH-sensitive moiety. At neutral condition, copolymer exist expanded form so that the carboxylic group of MAA tend to be ionized. In contrast, carboxylic group of MAA is unionized at acidic conditions, so copolymer exist shrinked form. Shrinking copolymer would give a stress to liposome, so permeability of liposome is increase. Role of immobilized GOx is inducing acidic pHs. GOx catalyzes the oxidation of beta-D-glucose into gluconic acid. When liposome were exposed in glucose exist environment, surrounding pH was decreased so that the liposome could be released. For this reason, the liposome can be applying drug delivery for type I diabetes. |
