Automated strain development using robotics and CRISPR genome engineering |

A biofoundry provides an integrated robotic platform to enable the rapid design, construction, and testing of genetically reprogrammed organisms for biotechnology applications. I present here the SKy Biofoundry to perform automated strain development using robotics and CRISPR genome engineering. The SKy Biofoundry is capable of performing automatic actions for various molecular and microbiological experiments such as gene cloning and colony selections using an integrated robotic and CRISPR-genetic platform. To facilitate the power of the automated platform, high fidelity CRISPR-base editing system was developed to create a sequence-specific cytosine conversion without the off-target mutations. The SKy Biofoundry has been hosting to construct massive genetic variants of Corynebacterium glutamicum and Escherichia coli using the CRISPR-Cas system to promote automated strain development in the Design-Build-Test-Learn cycle. |
