Fabrication of AB2 miktoarm copolymers micelles for anticancer drug delivery |

AB2 miktoarm copolymers which mimic structural characteristics of phospholipid are proposed as an alternative to linear copolymers. We synthesized a series of AB2 miktoarm copolymer consisting of PEG as A block and PCL as B block. All synthesized diblock copolymers with fPEG in the range of 0.50 ~ 0.17 formed spherical micelles when they self-assembled in aqueous solution. Contrastively, The PEG-PCL2 miktoarm copolymers with relatively long PCL blocks (fPEG = 0.32 ~ 0.15) formed worm-like micelles in aqueous solution. In the encapsulation study of doxorubicin (DOX) to evaluate their drug loading capacity, Worm-like micelles showed better loading capacity than spherical micelles. In vitro drug release test revealed that both of the DOX-loaded worm-like and spherical micelles displayed sustained drug release patterns, and worm-like micelles exhibited slower release rate than spherical micelles. PEG-PCL2 miktoarm nanocarriers exhibited enhanced cellular uptake into cells compared to DOX. |
