Cartilage regeneration using a biocompatible hydrogel in vivo |

Injectable hydrogels may serve as temporary scaffolds to guide cell attachment and differentiation of chondrocytes, resulting in newly formed cartilage tissue. Compared to preformed hydrogels, injectable hydrogels have various advantages. MPEG-PCL diblock copolymers consisting of methoxy polyethylene glycol (MPEG, 750 g/mol) and poly(ε-caprolactone) (PCL) is based on the idea that certain biomaterials undergo a simple liquid-to-gel phase transition under physiological conditions. This hydrogel constructs were then injected subcutaneously in nude mouse and they were retrieved after 1, 2, 4 weeks respectively. The composite hydrogel carried a property of sol–gel transition and thus formed a gel at body temperature. We demonstrated that gelation behavior of hydrogels cross-linked with primary chondrocytes can be controlled by regulating the specific interactions between the polymer and the cells. Hydrogels provided an appropriate environment for cartilage regeneration in vivo. |
