Neurogenic Differentiation of rat Bone marrow Mesenchymal Stem Cells using Small Compounds |

The aim of our research was thus to identify synthetic chemical molecules that might induce neuronal differentiation of BMSCs. During the course of this screening process, we identified the purine structure-based compounds. Here, we report the first evidence that N-cyclopentyl-N-(4-methoxybenzyl)-9-methyl-9H-purine-2,6- diamine (CMP) and N-(4-methoxybenzyl)-N-methyl-9H-purine-2,6-diamine (MMP) induce neurogenesis of rBMSCs. The larger proportion of NSE- and Tuj1-positive cells relative to CNPase-positive cells as shown by immunostaining suggests that rBMSCs differentiated mainly into neurons. These results support the concept that rBMSCs are potentially a rich source of neurons that can be induced to differentiate using chemical molecules based on familiar chemical series.
