Cross-linked Alginate Beads prepared by using Electrospinning for Adenovirus Delivery System |

In this study, ADV encapsulated alginate beads were fabricated successfully by using electrospinning method. Beads size gradually increased as the concentration of alginate and the flow rate increased. Crosslinking density of alginate beads increased with an increase of the concentration of calcium chloride. Encapsulation efficiency of alginate beads was characterized by using confocal laser microscopy. Release profile of the ADV encapsulated alginate beads in 37 °C PBS was obtained by photo-luminescence measurements. The ADV encapsulated alginate beads was injected onto the cancer cells. The ADV encapsulated alginate beads resulted in sustained release of ADV with slow rate for a long period due to biodegradable cross-linked alginate, which can maintain the treatment effects. This result was corroborated by FACS analysis. The ADV encapsulated alginate beads may be useful for delivering viral vectors into the cancer tissues. |
