3D on-chip flow-through polymerase chain reaction utilizing a single heater |
초록 |
Two poly(dimethylsiloxane) (PDMS) substrates patterned with microchannels were vertically stacked to realize a 3D fluidic conduit and the assembled microdevice was used for performing on-chip continuous-flow polymerase chain reaction (PCR). The assembled two PDMS substrates were bonded with a glass substrate and placed atop a heater controlled at the denaturation temperature of the nucleic acid. When the assembled microdevice is placed atop a heater with the glass-side down, the interface of two PDMS assembly displayed relatively lower temperature than that of the PDMS-glass assembly, as temperature gradient is formed in vertical direction due to the low thermal conductivity of the PDMS. A desired annealing/extension temperature was precisely controlled by molding the bottom PDMS layer to an appropriate thickness. In this way, multiple heating sources were replaced with a single heater, enhancing the device portability. |
