HPLC is essential to analyze the key ingredients of Korean Ginseng, ginsenosides, and also separate individual ginsenosides. Understanding the interaction between the carrier fluids and HPLC column adsorbents is important for the tasks of analysis and component separations. Gradient elution is an important technique in HPLC analysis. When two or more different eluants are mixed along the line of elution, we observe density changes and viscosity changes, which affects the pressure profiles. The pressure profile evolution is due to these rheological properties variation, and we use the pressure profile data to calculate the corresponding rheological data. This dispersion of multicomponent carrier fluids can show up both in pressure profile and light absorption profile. The relation between the pressure profile, aggregation structure characteristics and also diagnosis of column will be discussed together with the gradient elution conditions. |
