Development of Self-assembled Polymeric Nanoparticles for Drug Delivery Based on Hypoxia-triggered System |

Hypoxia is an important hallmark of cancer. Due to its unique feature that is rarely seen in normal tissue, hypoxia is being magnified as a primary target in the development of therapeutic drugs. In this study, we attempted to develop hypoxia-sensitive self-assembled nanoparticles which can selectively release the hydrophobic drugs under hypoxic conditions. To prepare hypoxia-responsive nanoparticles (HR-NPs), a hydrophobic 2-nitroimidazole (NI) derivative was conjugated to the backbone of hydrophilic carboxymethyl-dextran. Due to the property of hypoxia-responsive NI derivative, the drug release rate of HR-NPs remarkably increased under the hypoxic condition. In addition, HR-NPs were selectively accumulated at the hypoxic tumor tissues after systemic administration into the tumor-bearing mice. Overall, the HR-NPs might have a potential as the drug carrier for treatments of hypoxia-associated diseases. |
