The Inflammatory Responses to PLGA Depending on Hydroxybenzyl Alchohol : In Vitro and In Vivo Test |

PLGA has demonstrated satisfactory biocompatibility, although some reduction in cell proliferation and transient tissue inflammation has been reported. p-hydroxybenzyl alcohol (HBA) is one of phenolic compounds in herbal agents and plays a pivotal role in protection against oxidative damage-related diseases due to anti-inflammatory effects. We have developed HBA-incorporated peroxalate polymer (HPOX). The cytotoxicity of HPOX in PLGA was investigated using fibroblasts by a MTT reduction assay. The anti-inflammatory effects of HPOX on inflammatory response of LPS-treated macrophage cells was evaluated by measuring the level of tumor necrosis factor-alpha (TNF-a). The PLGA/HPOX films had no adverse effect on fibroblasts cell viability and elicited significantly lower production of TNF-a than PLGA films. To further demonstrate in vivo inflammatory response surrounding the implants, histological examination was performed. This research was supported by WCU(R31-20029) |
