Gamma-valerolactone (GVL) is an organic compound with the formula C5H8O2. Its precursor 4-hydroxyvaleric acid (4HV) is one of the industrial important hydroxyl acids used to produce various commodity chemicals and biofuels and can be polymerized into biodegradable polyesters. In the present study, we employed Escherichia coli and Pseudomonas putida strains to produce 4HV from levulinic acid (LA). In E. coli, 4HV production was performed by expressing 3-hydroxybutyrate dehydrogenase (3HBDH) and whole cell biotransformation from LA, resulting in 7 g/L of 4HV production in glucose-fed anaerobic environment. We have engineered P. putida strain to block the substrate degradation by its native degradation pathway. The resultant strain produced 15 g/L of 4HV from 20 g/L of LA in 48h of cultivation. |
