Biomimetic Hierarchical Structures of Collagen-like Peptide Amphiphiles |

Hierarchically structured biomaterials are finding widespread applications in regenerative medicines. Despite advances in material synthesis and assembling processes, it is still challenging to precisely control the hierarchical assembly. Here, we demonstrate a biomimetic approach to create collagen-like hierarchical materials. We first synthesized a collagen-like peptide amphiphile identified by phage display, which features collagen type I's characteristic tri-peptide repeating unit, complemented by a hydrophobic tail to stabilize the nanofibrous structures in molecular self-assembly. Then, we assembled the amphiphiles into hierarchically organized matrix by local accumulation at meniscus on a substrate, which leads to deposition of liquid crystalline structures. The resulting structures provided oriented growth of bone cells and biomineralization of hydroxyapatite. We believe that the hierarchical collagen-like materials can be used for the replacement of damaged tendon tissues and the artificial bone. |
