The transplantation of RPE has recently become a possible therapeutic modality for retinal degeneration. To evaluate usefulness of silk as substrates for RPE transplantation, we fabricated thin silk films. RPE cells were seeded on TCP, PLGA films and silk films. We measured the cell adhesion, cell viability, morphology and specific mRNA expression by MTT assay, SEM, Immunofluorescence and RT-PCR. In this study, we confirmed that the cell viability and proliferation were slicely different between silk and PLGA films. But, silk is an attractive material because of natural. The phenotype of the cells was maintained in silk films. The RT-PCR results showed specific mRNA expression. In conclusion, this study suggested that silk films may serve as potential RPE cells' differentiation and structural basis for tissue engineered RPE cells.
