Antibody-based conjugates have become an essential component in a variety of applications from immunoassays to drug conjugates because of their proven efficacy and selectivity to target. However, it is obvious that properties and functions of the conjugates can be influenced by the site at which the antibody is modified. Thus, approaches to conjugate molecules to IgG site-specifically have been developed, in particular for application to therapeutics. In this presentation, we describe our strategy to conjugate IgG and protein using photo-reactive amino acids. To enhance the reactivity and site-specificity of the analogues, the unnatural amino acids were installed in an Fc-binding peptide. The conjugation reaction was optimized, and a purification procedure was established. The developed method was applied to conjugate several proteins to IgG, and the biochemical properties and biological functions of these conjugates were characterized. |
