제목 BME Release Pattern from β-mercaptoethanol/ Poly(lactic-co-glycolic acid)(BME/PLGA) Scaffolds and Neurogenesis of Bone Marrow Stromal Cells
초록 PLGA is a biodegradable polymer with acceptable mechanical strength and well-controlled degradation rate. We fabricated BME/PLGA scaffoldsfor neurogenesis of BMSCs and to observe the release behavior of BME. BME/PLGA scaffold were manufactured as the concentrations of 200,400,600μM,1mM of BME by solvent casting/salt leaching method. Cellular viability and proliferation were assayed by MTT test. Morphology were confirmed by SEM. The effect of BME/PLGA scaffold on the neural differentiation of BMSCs were assessed RT-PCR. The release profiles of BME were assayed using UVspectrometer. Results, BME of 600 μM loaded scaffold revealed low cell viability in the results of MTT. Mophology of BMSCs changed like a neural cell in BME of 200,400μM loaded PLGA scaffold. BME was sustainly released from BME/PLGA scaffolds for 30days. We conclude that neural differentiation of BMSCs in BME/PLGA scaffold could be possible for long period.
