Engineered magnetic nanoclusters for sensitive diagnosis of atherosclerosis via magnetic resonance imaging |

Oleyl dextran-coated magnetic nanoclusters (ODMCs) were fabricated for magnetic resonance (MR) imaging of atherosclerotic plaques. We synthesized ODMCs by enveloping magnetic nanocrystals (MNCs) as MR probes with conjugates of hydrophobic oleic acid and hydrophilic dextran, which is known to specifically bind to macrophages through scavenger receptors (ODex). The magnetic properties of ODMCs were engineered by control degree of substitution (DS) of oleyl group in ODex and the concentration of ODex used for coating of MNCs. Engineered ODMCs exhibited high MR sensitivity and efficient targeting ability against macrophages without cytotoxicity. In vivo MR imaging after intravenous injection of ODMCs into a rat balloon injury model showed considerable decrease of MR signal intensity at plaques of the injured carotid artery. From these results, we verified that ODMCs have sufficient potential as a MR imaging agent for diagnosis of atherosclerosis. |
