Dual Growth Factor-Loaded Porous PCL Scaffolds for Chondrogenesis of Adipose-Derived Mesenchymal Stem Cells |

It is well known that polymeric scaffolds with controlled release of growth factors (GF) may be effective for tissues regeneration. Based on the high specific binding of heparin for various GFs, several heparin-containing systems have been developed for the controlled release of GFs. In this study, we prepared polycaprolactone (PCL)/Pluronic F127 scaffolds which can bind with heparin (by hydrogen bonding between the heparin and Pluronic F127 exposed onto surfaces) and the following GFs. To induce the chondrogenesis of adipose-derived mesenchymal stem cells (ADMSCs), TGF-β2, BMP-7 (single GF system) and TGF-β2/BMP-7 (dual GF system) were introduced onto the PCL/F127 scaffolds, respectively. Their release behavior was quantified by ELISA kit. The chondrogenesis (in vitro) of ADMSCs in scaffolds was also investigated by histological and molecular biological analyses (This work was supported by a grant from Korea Science & Engineering Foundation: Grant No. R01-2006-000-10432-0). |
