In order to improve the solubility of nifedipine (NIF) and improve the release rate of NIF, the novel mold device was used in this system. The squeeze-osmotic pump tablet system (SQT) was composed of three cylindrical squeeze-push layers (A,B,C) and drug located in center of tablet, the materials of push layer A and C were the same, however, push layer B was different from A and C in formulation. Tablet was coated with cellulose acetate membrane (CA). TiO2 as a white pigment was added into CA membrane to prevent light degradation of NIF. In addition, two orifices were on both side surfaces. During the release test, a 1-ml of water phase (Na2HPO4, SLS and citric acid monohydrate) (pH = 6.8) as a release medium was pipetted out and kept in an amber auto-sampler vial (1, 4, 8, 12, 14, and 24 hrs). All the samples were analyzed with HPLC system within 1 day. It showed that SQT using novel mold device could greatly improve solubility and release rate of NIF. |
