Characterization of an alpha-mannosidase from Streptomyces coelicolor |

Recently, glycosylation of specific proteins in prokaryotes have gained much attention due to their novel role in control over various biological pathways. We analyzed the role of an alpha-mannosidase from Streptomyces coelicolor M145 and have shown a possible role of this protein in regulation of antibiotics production. α-mannosidase was found to be the single ORF annotated to encode an alpha-mannosidase (AM1) from S. coelicolor M145 and was well overexpressed in Escherichia coli BL21(DE3). Purified recombinant AM1 of 110 kDa exhibited alpha-mannosidase activity and a deletion mutant for α-mannosidase exhibited interesting growth phenotype depending on the carbon source in growth media. The results obtained emphasize that this protein could possibly involve in de-glycosylation of specific protein(s) that influence actinorhodin (ACT) and undecylprodigiosin (RED) in Streptomyces coelicolor. |
