A simple Liquid Chromatographic methode for the determination of 'Glimepiride'. 20㎕ of 2ppm Glimepiride the filtrate was injected to extraction column(Capcellpak MF ph coulumn 4.0 × 10 mm) with mobile phase A, 6.4mM sodium phosphate : Acetonitrile= 50 : 50 (pH 2.7) dilution to 2times in water. After concentration with intermediate column(Capcellpak C18 UG120 S5 2.0 × 35mm), valve A was switched to valve B in order to inject analyt to analytical column(XTerra C8 3.9 × 150mm) with mobile phase B, 6.08mM sodium phosphate: Acetonitrile= 47.5 : 52.5 (pH 2.7) and UV detector at 228㎚. Linearity and precision were checked over the concentration range 10ng/㎖～1000ng/㎖. Limit of detection was 5㎍/㎖. |
