제목 | Sustained release of nifedipine from multi-coated granules prepared by porous membrane and evaluation of in vitro release test |
초록 | Among the various novel drug delivery systems available in pharmaceutical industry, oral delivery systems hold the major part because of their obvious advantages of ease administration and better patient compliance. In order to prepare new device of granules contained water-insoluble drug for oral administration and improvement of release pattern. We prepared osmotic multi-coated granules by fluidized-bed spray coating method, and used nifedipine (ND) as a model-drug. The osmotic multi-coated granules composed of sugar, various excipients, such as hydroxypropyl cellulose, lactose, fructose, cellulose acetate, and Eudragit® has been successfully developed with purpose of delivering ND. The present study is intended as an investigation of the effect of different preparation factors, such as bead sizes, membrane thickness and porosity, coating process, and excipients to achieve optimal release performance. Based on the results of in vitro release test, release rate of ND granules were associated with membrane thickness, bead sizes due to the small granules has been exhibited faster release rate than the large, porosity, and the amount of excipients such as disintegrant and binder. In conclusion, we confirmed that our device sustained a release in 24 hours with near zero-order release. Acknowledgement: This work was supported by KMOCIE |
