Preparation of theragnostic agent for multifunctional drug delivery system |

The poly amino acid, PHEA(Poly-hydroxyethylaspatamide) is known as a useful biomaterial with appropriate structural modification. In this study, we synthesized PHEA-mPEG-C16 by condensation polymerization and synthesized with iron oxide by coprecipitation method for using a contrast agent. It called PSI-USPIO, we loaded hydrophobic drug such as doxorubicin into the PSI-USPIO. Finally, we were prepared mutifunctional drug deliver carrier which combined system of imaging and therapy whole in one and called PSI-USPIO-DOX. We were characterized physical property by ELS, TEM, ICP analysis. The intracellular uptake was confirmed against B16F10 murine melanoma cell by fluorescence activated cell sorter(FACS) and conforcal laser microscopy. The anticancer activity was confirmed in mice bearing of B16F10 murine melanoma model as growth pattern of the tumor and observed in rabbits bearing of VX2 hepatoma model by 3.0T clinical magnetic resonance image (MRI) system. |
