A dTDP-4–keto–6–deoxy–D-glucose is a key intermediate in antibiotics such as aminoglycosides, polyketides and macrolides. A novel enzymatic synthesis of dTDP-4-keto-6-deoxy-D-glucose, starting from dTMP, acetylphosphate and glucose-1-phosphate, was produced with four enzymes. For the efficient production, Recombinant E.coli strains containing TMP kinase, acetate kinase, dTDP-glucose synthase and dTDP-D-glucose 4,6-dehydratase were used for one-pot synthesis. Recently, two enzymatic synthesis using dTDP-glucose synthase and dTDP-D-glucose 4,6-dehydratase were reported. However, our proposed synthetic method replaced expensive dTTP with a cheaper dTMP in point of economical production. To implement proposed reaction scheme, TMP kinase and acetate kinase are additionally incorporated into the dTDP-glucose synthase and dTDP-D-glucose 4,6-dehydratase. Finally, dTDP-4-keto-6-deoxy-D-glucose was synthesized using the four enzymes in one-pot reaction from dTMP and glucose-1-phosphate. |
