Co-culture according to the Ratio of OECs and BMSCs on Differentiation and Proliferation of BMSCs |

OECs play an important role as they are the source of different trophic substances and present a great neurotrophic activity. BMSCs are important progenitor and supporting cells that have the intrinsic ability to self-renewal and differentiate into multiple functional cells. We have investigated the differentiation of BMSCs through co-culture system with OECs. Expression of the GFAP, S-100, P75 and NF were determined by staining. RT-PCR was conducted to confirm mRNA expression of NF, P75 and BDNF in BMSCs co-cultured with a high percentage of OECs. Following co-culture time with a high percentage of OECs, BMSCs underwent morphological changes similar to those of cultured OECs and stained positively for GFAP, S-100, P75 and NF protein as well as high proliferate aspect were detected. These findings support the assertion that BMSCs can be differentiated into cells that are OEC-like in terms of phenotype using OEC only.
