The reconstructed human skin equivalents have been widely studied to improve the structural and functional integrity in various methods. In this study, we present a new strategy to introduce keratinocyte monolayer in reconstructed human skin equivalents via cell sheet engineering. We used poly(N-isopropylacrylamide)(pNIPAM), a temperature-responsive molecule, onto a conventional tissue culture polystyrene plate to fabricate the temperature-responsive culture surface. Cultured keratinocyte monolayer sheet could be harvested as an intact cell sheet by reducing the culture temperature. The cell sheet was transplanted and submerge-cultured on the dermis layer, and then differentiated to cornified layer with air-lift culture. Overall, this strategy to fabricate the 3D reconstructed skin using cell sheet engineering would be used in tissue engineering, drug delivery, and cosmetics assessments. |
