Magnetite nanoparticles in microsphere for intestinal delivery |

Magnetite nanoparticles are degraded in acidic solutions, therefore cannot be employed for peroral delivery. Amphiphilic polymer-stabilized magnetite nanoparticles were prepared for efficient encapsulation within pH-sensitive microspheres. A di-block copolymer composed of PCL and methoxy PEG (mPEG-PCL) was dissolved in dichloromethane and FeCl₂.FeCl₃ in aqueous phase was added to organic phase. Ammonium hydroxide solution was added to mixture with the block copolymer. Eudragit L100-55 and the magnetite nano-aggregates in a methanol/acetone mixture were poured in mineral oil for W/O emulsification. The magnetic properties of the nano-aggregates were confirmed by vibrating sample magnetometer (VSM), suggesting that incorporation of mPEG-PCL slowly decreased T2-relaxation time. Dynamic light scattering (DLS) showed incorporation of mPEG-PCL decreased particle sizes. Encapsulation efficiencies of the magnetite nano-aggregate were also affected by the amount of mPEG-PCL in the aggregates. |
