Hybrid system composed of baculovirus and Galactosylated PEI for efficient gene carrier |

Baculovirus has been developed as an attractive viral vector system due to its advantages such as safety, large cloning capacity, the lack of replication and preexisting immunity. However, baculovirus system has several drawbacks. Especially, to enhance the efficacy of gene delivery, lack of specificity and the inactivation of baculovirus by the complement system in human serum should be overcome. Therefore, hybrid system (GP/B) was constructed by electrostatic interaction between baculovirus (B) and galactosylated polyethylenimine (GP) in this study. The resulting GP/B had the positive aspects of both viral and non-viral vector systems. Furthermore, the GP/B significantly enhanced transfection efficiency and showed good cell-specificity compared to either viral or non-viral vector systems. These results suggest that the GP/B hybrid system can be used in gene therapy to enhance transfection efficiency and hepatocyte specificity. |
