제목 Selective stimulation of caveolae-mediated endocytosis and hepatocyte-targeting by an osmotic polylactitol-based gene transporter
초록 Here, a new multifunctional polymeric nanocarrier, polylactitol-based gene transporter (PLT) is developed by crosslinking low molecular weight branched polyethylenimine (b-PEI) with lactitol diacrylate. Polylactitol has multifunctional properties having three key functions for successful gene delivery: 1) hyperosmotic part trigger caveolae-mediated endocytosis, 2) galactose part can deliver genes into the hepatocyte via receptor-mediated endocytosis, and 3) PEI part assists rapid endosomal escape of gene due to proton sponge effect. As expected, PLT/DNA nanocomplexes showed low cytotoxicity, significant transfection efficiency, hepatocyte-specific targeting, and transition of cellular uptake pathway into caveolae-mediated endocytosis avoiding lysosomal degradation. Our results suggested that polylactitol-based gene transporter (PLT) is safe and efficacious vector, which will highlight its potential in the treatment of hepatic diseases.
