Preparation of Tramadol-HCl Loaded Microsphere for Control Release |

Tramadol-HCl (TH) is a weak μ-opioid receptor agonist and a reuptakes inhibitor of serotonin and of norepinephrine. Hydrophobic polyoxalate (POX) has been suggested for use in biodegradable medical devices such as H2O2-sensitive bioimaging probe and controlled release formulation. In this study, we developed tramadol-encapsulated POX microspheres by changing molecular weight of the POX and methods to prepare microsphere including W1/O/W2, O1/O2/W, O/W, and O1/O2. The inner and outer morphology and physicochemical properties of TH-encapsulated POX microspheres was characterized using SEM, XRD, DSC, FT-IR and HPLC. Through the study results, it shows that the source of the skin layer of TH-loaded microspheres is comprised of POX polymer and TH was encapsulated within the microsphere.
