Cyclodextrins (CD) are macrocyclic oligosacharides, containing 6(α-CD), 7(β-CD), or 8(γ-CD) D-glucose units. CD can be used to improve solubility and bioavailability of poorly water soluble drugs (ID) as well as other properties such as stability, dissolution rate, and etc. Kollidone® is one of water-soluble polymers widely used as a pharmaceutical excipient. In this study, the inclusion complexes of ID/CD/Kollidone® were prepared by vortexed-mixing, kneading, rotary evaporation, co-precipitaion or spray-drying method. The physicochemical properties of ID/CD/Kollidone® inclusion complexes were characterized by scanning electron microscopy (SEM) and powder X-ray diffraction (XRD). SEM observation of ID/CD/Kollidone® inclusion complexes prepared by spray drying method revealed that they were spherical. Significant reduction of the crystallinity of ID/CD inclusion complexes incoporated in Kollidone® was observed by XRD. The solubility and dissolution rate of ID were significantly improved by complextion with CD and Kollidone® with reproducibility. These results suggested that inclusion complexes could be used as a pharmaceutical formulation for clinical application of ID. |
