Synthesis and Characterization of Mannose-PEG-linked Superaramagnetic Iron Oxide Nanoparticles specifically targeted to antigen presenting cells for MR imaging on Lymph Node |

The aim of the study is to prepare biocompatible and specifically targetable nanoparticles to lymph nodes (LN) for LN specific MRI. Previously many contrast agents have been developed to target to antigen-presenting cells for LN detection by MRI. In this study, mannose-PEG was prepared from D-mannopyranosylphenyl isothiocyanate and Amine PEG Carboxyl. The APTES-activated SPION and mannose-PEG was crosslinked to produce mannose-PEG-linked SPION. Mannose on the surface achieved efficient targeting to mannose receptor on antigen presenting cells. PEG corona layer prevented from aggregation during the systemic circulation with accompanying enhanced specificity and minimized systemic toxicity. In in vivo study, rats were intravenously injected with Mannose-PEG-SPION and tracked by MR imaging. MR imaging on LN clearly revealed the preferential uptake of Mannose-PEG-SPION in immune cells-rich lymph nodes. The accumulation in LN was also confirmed by Prussian blue staining. |
