Formation of misfolded proteins aggregates called amyloid fibrils, and the intermediate state of amyloid fibrils is regarded as fatal to patients because of their physical properties such as insoluble protease resistance. However, those properties could also make them useful to create new nano-materials. Amyloid fibrils formed as such are highly ordered, long, and unbranched threads with robust mechanical strength but still with flexibility. In present study, we propose a novel strategy to fabricate unique multilayer thin films based on amyloid fibrils of κ-casein which is one class of caseins in bovine milk and has the ability to form amyloid fibrils under certain conditions. Prior to prepare amyloid-based multilayer films, the formation rate of κ-casein fibrils was monitored under different temperature and pH conditions with thioflavin-T assay. The amyloid multilayer thin films were prepared by the sequential deposition of κ-casein fibrils and positively charged polyelectrolytes. |
