Osteogenic Differentiation of Human Adipose-Derived Stem Cells Is Promoted Through Remodeling of Type I Collagen by MMP-13 |

Matrix metalloproteinases (MMPs) has the essential role for the cell biology through cleavage of extracellular matrix (ECM) proteins. Bone tissue is mainly composed of type I collagen (ColI), which is known to promote osteogenic differentiation of human adipose-derived stem cells (hASCs). However, still not clear that the mechanism of enhancing osteogenic differentiation by ColI. It has been proven that cell-matrix interaction plays an important role in osteogenic differentiation of hASCs. MMP-13 is significantly increased in hASCs during osteogenic differentiation on ColI. After osteogenic induction, the mRNA expression levels of both ITGA3 and RUNX2 increased significantly on ColI-coated plates. MMP-13 has a role as a regulatory factor in bone formation, which is regulated by transcription factor RUNX2. These results collectively suggest that hASCs-ColI interaction can enhance osteogenic differentiation through ColI/MMP-13 positive feedback loop. |
