Regeneration of Biodisc Using Annulus Fibrosus Cell on Hyaluronic Acid Loaded PLGA Scaffold, in vivo |

Poly(lactide-co-glycolide)(PLGA) and hyaluronic acid (HA) has been widely used as biocompatible scaffold materials to regenerate tissue. In this present study, we fabricated microporous PLGA and HA loaded PLGA scaffolds by salt-leaching and then penetrating method. AF cells were seeded in HA/PLGA scaffold, and then PLGA and HA/PLGA for the in vivo study were implated in 4 week old nude mice. All scaffolds were characterized using a scanning electron microscope(SEM) / Reverse transcription polymerase chain reaction(RT-PCR) was assessed to measure mRNA expression and glycosaminoglycan(GAG) quantity from annulus fibrosus cells in scaffolds. It was proven by higher specific mRNA expression in the HA-PLGA scaffold than PLGA alone. In addition, annulus fibrosus strongly produced sGAG in HA/PLGA. This result indicates that HA/PLGA scaffold is useful for intervertebral disc regeneration . This research was supported by WCU(R31-20029) |
