Cloning and expression of human coagulation factor VII |

Coagulation factor VII is involved in the initiation of blood coagulation by forming the complex with tissue factor, which catalyzes thrombin generation and clot formation. The isolation and purification of FVII from available species is very challenging because it is present in extremely low levels in plasma. A cDNA encoding FVII was isolated by reverse transcriptase-mediated DNA amplification using a human liver cDNA library. The recombinant FVII was cloned and expressed using Pichia expression system. The expressed factor may be an important new tool in the development of novel antithrombotic drugs. |
