Here we report on the fluorescence resonance energy transfer (FRET) between chromophores assembled with DNA as a template. Single strand DNA molecules conjugated with three chromophores, JOE, TAMRA, and TexasRed, are self-assembled with its complementary matrix DNA strand. The FRET efficiencies for the single step energy transfer of JOE-to-TAMRA and TAMRA-to-TexasRed pairs are 29.77 % and 37.24 %, respectively, in aqueous milieu at 4 °C. Above the melting temperature of the DNA duplexes used, no significant FRET is observed. Two-step FRET of JOE-to-TAMRA-to-TexasRed shows 21.95 % FRET efficiency. This study suggests that DNA can be used as a versatile platform for the precise assembly of chromophores to facilitate the energy transfer.
