제목 Functionalized nanoparticles as adjuvant carrier for dendritic cell-based cancer immunotherapy
초록 Dendritic (DCs) are potent professional antigen presenting cells capable of initiating a primary immune response and activating T cells and which a pivotal role in host immune response for cancer. Prior to this process, effective antigen and adjuvant should be delivered in DCs to induce maturation and cytokine generation. Here, we developed poly(lactic-co-glycolic acid) (PLGA)-NPs encapsulated with OVA and poly IC to increase uptake, leading that NPs prompt DCs maturation. PLGA-NPs showed efficiently taken up by DCs and greatly facilitated MHC I antigen presentation. Moreover, vaccination of DCs treated with PLGA (OVA+poly IC)-NPs leads to generation of OVA-specific CD8 T cells, resulting antitumor efficacy was significantly increased in EG7 tumor bearing mice compared to control (p < 0.001). Taken together, we demonstrate that PLGA-NPs system were useful strategy to deliver tumor specific antigen or adjuvant into DCs, which lead to application for DCs-based cancer immunotherapy.
