As DNA molecules are the support for the genetic information, the information of their sequence is very important from many points of view. With the advent of PCR, it is now possible to exploit the sequence information from DNA. Despite the success of conventional sequencing, the quest for alternative methods is an active field of research. Recently various single molecule manipulation techniques have been developed, allowing a direct investigation of DNA mechanics. In this study, two ends of dsDNA were pulled apart along the opposite direction under constant velocity or constant force by using molecular dynamics. When the unzipping forces F(z) were measured as a function of extension z, it reveals that the curve shows several maxima and minima. It has been found that this pattern is closely related with the DNA sequence. It is concluded that unzipping experiment of dsDNA may provide an alternative method to determine DNA sequence without any other aids of enzymes or complex devices. |
