Small interfering RNA (siRNA) is known as promising therapeutic agents for the post-transcriptional gene silencing with a highly sequence-specific manner. As a novel strategy to enhance the delivery efficiency of siRNA, we increased the charge density of siRNAs by hybridization between dimerized or trimerized single strand of sense and antisense siRNAs. By using dimerized sense/antisense siRNAs and trimerized sense/antisense siRNAs, multiple siRNA conjugates were prepared as a linear form and dendrimeric form, respectively. Mesh-like structure of dendrimeric multi-siRNAs was confirmed by AFM and confocal microscopic image. With a cationic polymer as a delivery carrier, these linear and dendrimeric multi-siRNA conjugates were complexed as a nano-sized particles and delivered into the cancer cells to silence the target gene expression. As a result, dendrimeric multi-siRNA conjugates showed more enhanced target gene inhibition than linear multi-siRNA conjugates or naked siRNAs. |
