제목 A MALDI-MS-based quantitative analytical method for endogenous estrone in human breast cancer cells
초록 In humans, the level of endogenous estrone is highly correlated with postmenopausal breast cancer incidence. However, it is difficult to quantitatively monitor endogenous estrone due to its low concentration. In this study, we develop a highly sensitive quantitative platform based on mass-assisted laser desorption/ionization mass spectrometry (MALDI-MS) to analyze the absolute amount of endogenous estrones in various clinical specimens. Through the modification of the endogenous estrone, it achieved excellent linearity (R2> 0.99) and increased platform sensitivity (quantitation limit: 11 fmol). In addition, we were able to confirm the absolute amount of endogenous estrone using deuterated estrone as an internal standard in cells of the breast cancer cell line MCF-7 (34 fmol / 106 cells). Finally, through the MALDI-MS-based quantitative platform, the changes in the levels of metabolic expression of estrones (17.7 fmol/106 letrozole-treated cells) in MCF-7 cells treated with aromatase inhibitors were successfully monitored.
