Chemical modification of chitosan with 2-aziridineethanol was carried out in the presence of aqueous hydrochloric acid for the increase of amine value. The molecular weight and chemistry of the reacted chitosan were characterized using GPC, 13C and 1H NMR, respectively. The results indicated that the amines of chitosan was reacted with 2-aziridineethanol to give the amine values increased chitosan, which was controlled by the feed molar ratio of 2-aziridineethanol/amine in chitosan. Gel electrophores is showed that DNA migration was retarded completely at a N/P ratio of 1/2, indicating good DNA condensation capability of the amine values increased chitosan. The amine values increased chitosan showed a comparable cytotoxicity with those of chitosan itself. Gene transfection efficiency of the amine values increased chitosan will be also discussed. |
