제목 Transplantation of 3D-Printed Tracheal Graft Combined with iPS Cellsin Segmental Tracheal Defect Model
초록 Here, we designed a artificial trachea which can enhance the regeneration of tracheal mucosa and cartilage through the optimal combination of 3D-printed scaffold and iPS-derived cells. Human bronchial epithelia cells (hBEC), iPS-mesenchymal stem cell (iPS-MSC) and iPS-chondrocytes (iPS-Chd) were used for regeneration of tracheal mucosa and cartilage in vivo. Each cells were seeded onto a surface of inner (hBEC) and outer layer (iPS-MSC or iPS-Chd) of scaffold. It was then transplanted in a segmental trachea defect. After 4 weeks postoperatively. Endoscopy did not reveal granulation ingrowth into tracheal lumen. Alcian blue staining clearly showed formation of ciliated epithelium in iPS-MSC groups. In addition, Micro-CT analysis proved that iPS-Chd groups were the most effective in forming neocartilage in defect sites. Therefore, this study provided a promising approach for the 3D reconstruction of a segment tracheal defect.
