CuAAC(click) reaction was well-known to transform 1,4-triazole between alkyne and azide. In this study, we synthesized two fluorescent peptidyl probe (1, 2) for monitoring intramolecular CuAAC reaction in 1% aqueous solution. These synthesized two kinds of probes also showed the effect of Arg amino acid on aqueous conditions. Upon addition of Cu(II) in the presence of sodium ascorbate, intramolecular CuAAC reaction of the probe occurred, resulting in the enhancement of monomer emission at 400 nm and decrease of excimer emission at 480 nm. As the concentration of Cu(II) increased from 0.5 to 3 equivalent, the rate of click reaction increased. We investigated the rate of CuAAC reaction controlled by equivalent of Cu(II) and the reaction rate was proportion to the concentration of Cu(II) in HEPES or phosphate buffered solutions at pH 7.4. |
