As a metabolic engineering strategy, adaptive laboratory evolution (ALE) is often used to improve production by allowing cells to adapt to the culture conditions. Recent evolution technologies have enabled microorganisms to acquire desired biological traits at the laboratory level, faster than natural evolution. We designed a genetic mutator device that accelerated mutation rate and a genetic selector device that sorted out cells with a superior phenotype. We created various mutator devices exhibiting different mutation rates. We also created several selector devices responsible for different concentrations of a target molecule. Using the mutator and selector devices in conjunction with the genetic toggle switch, we were able to switch from the mutation state to the selection state simultaneously and vice versa. This genetic circuit will be useful to accelerate mutations and to screen cells having a desired biological trait, leading to the faster engineering of microorganisms. |
