제목 | Adeno-associated Viral Vector (AAV) System for the Treatment of Hypoxia Ischemic Encephalopathy (HIE) Brain Injury |
초록 | AAV has been widely used in various research both in vitro and in vivo, as well as in clinical trials for the development of gene therapy products. Several renowned AAV-based gene therapy products include Glybera, Zolgensma, and Luxturna. However, one of the main hurdles in commercializing gene therapy products is their cost. The high dose of AAV needed may be the main reason for the costly manufacturing of the gene therapy product. Therefore, the efficacy and safety issues of AAV are the main concerns when developing a novel product. Herein, we aimed to enhance the transduction efficiency of AAV by treating it with biomimetic phenolic molecules. Previously reported to have promoted the transduction towards the heart, we now demonstrate that the phenolic-AAV promoted the local transduction in HIE injured brain. The effect of the phenolic molecules in the transduction efficiency of AAV towards damaged brain regions may benefit the development of a highly potent therapeutic product for the treatment of numerous central nervous system (CNS) diseases.
