Efficient delivery of siRNA is a prime requirement of RNA interference (RNAi)-based cancer therapy. Here, we delivered Akt1 siRNA using biodegradable poly (ß-amino ester) (PAE) composed of low molecular weight polyethylenimine and poly (ethylene glycol) for lung cancer studies in vitro and in vivo. The silencing achieved with PAE carrier was ~1.5 times superior and safer than standard PEI25K. PAE delivered Akt1 siRNA silenced Akt1 protein and hindered cancer cell growth in Akt1 specific manner. It promoted Akt1 knock-down specific apoptosis and also reduced basic cancer properties like cell proliferation, malignancy and metastasis. Moreover, aerosol delivered PAE/Akt1 siRNA complexes in K-rasLA1 and urethane-induced lung cancer model mice efficiently suppressed lung cancer progression through regulating proteins important for Akt-related signals, and cell cycle regulation. The results of our study strongly highlight the potential of PAE delivered Akt1 siRNA in lung cancer treatment. |
