제목 Engineering aglycosylated antibody Fc for prolonged serum half-life
초록 Antibody Fc region is critical to prolong serum persistence of IgG antibodies through the pH-dependent FcRn binding and subsequent intracellular trafficking-mediated recycling mechanism In a production standpoint, aglycosylated antibodies, which allow simple bacterial expression, bypass glycan heterogeneity issue, and lower capital investment for production, has many advantages compared to conventional glycosylated antibodies produced in mammalian cells. We have engineered the aglycosylated Fc region for improved pH-dependent FcRn binding and the binding characteristics of the resulting aglycosylated Fc variants will be discussed.
