Here, we made self-assembled polymeric micelle immunomodulators using amphipathic polymers for cancer treatment. The cationic amphipathic polymer was obtained by synthesizing hydrophobic all-trans-retinoic acid and hydrophilic PEI. The cationic amphipathic polymer can self-assemble in water and form micelles with strong positive charge and proper particle size. These micelles exhibited enhanced cancer activity against drug resistance. We showed that the mechanical force generated by the cationic charge of micelles mainly induces apoptosis. Beside the cell lysate by necrotic cell death caused an antitumor immune response. In addition, these micelles were shielded by biocompatible polymers to enhance tumor selectivity. Finally, biocompatible polymeric shield micelles (BPM) accumulate selectively at tumor following systemic administration to tumor-bearing mice, resulting in anti-tumor immunity. Therefore, this new technology demonstrates great potential in clinical cancer immunotherapy. |
