Grandirubrine, a member of the unique family of naturally occurring tropoloisoquinoline alkaloids exhibits significant cytotoxic activity (IC50 = 0.53 μM against the leukemia P388 cell line). Its potent biological activity and challenging structure attracts a lot of attention. Recently we have reported the synthetic strategy of the radical anionic coupling reaction of phenolic nitronate for pareitropone, one of the tropoloisoquinoline alkaloids, and herein we newly report a successful application of the radical anionic coupling reaction to the grandirubrine synthesis.
